Are you riding with a back pack ? if the saddle setup is of it will agravate the situation because of the extra weight ( on your ass ) . dont worry , you will get use to it and it will ease.

If your saddle isn't set up correctly, then: yes, and no.

Is your saddle perfectly horizontal or pointing upwards?

My saddle is perfectly horizontal.

The pain has since subsided *weird*, as long as I know my nuts aren't going to fall off I'll stand the pain.

dude , they wont just fall off , you will need to have them surgically removed if the blood supply is stopped for too long ( you will know this if the pain gets worse and worse till you puke and cant walk )

But, if it's numb (from pinched nerves while on the saddle),

will I notice if the blood supply has been constricted?

Pinched nerve = numb / pins and needles feeling

Loss of blood for a short while = pain that will subside

Loss of blood and twist of blood vessel = horrific pain / loss of nut !! ( this is not too common though )

Dont stress , get the saddle setup checked .
