I'm going to the Eiland (ATKV - near Tzaneen) for holiday in December.

Which bike should I take with? (Road or MTB)

Any suggestions on routes / trails?

If you can, take both. I see they have a MTB trail but I don’t know anybody who did it. There are excellent MTB trails in the Magoebaskloof area about 100km from Eiland. Tzaneen Cycling also have a club ride every Saturday morning starting at 06h00 in Tzaneen (and sometimes at Eiland). PM me should you need more detail and I can keep you informed.
