Depends on two factors:

 

1. Size of hole

2. Size of bank balance

 

If 1 > 2, plug it. If that doesn't work, see above. If that doesn't work, replace.

Definitely depends on the damage done buy the puncture, if it's small enough it can definitely be plugged, however there are factors at play.

yes - just ream out the hole a bit before you put the new plug in

The plugs we use on tubeless bicycle tyres (mountain bike tyres anyway) are effectively identical to those used for roadside or bush repairs on four-wheel drives (diameter of the plug is just a little smaller). While I am quite capable of removing a tyre from the rim in the middle of nowhere and plugging it from the inside with a proper plug, it's not necessary unless the hole is pretty big. The external plugs work pretty well (sometimes two or three in a hole) and I have driven many thousands of kilometers without problems on tyres plugged with external plugs. My advice is plug it and forget it, especially if you have a decent sealant in your tyres. You really shouldn't have a problem – in fact you shouldn't even notice it. I have never had a problem with a plugged bicycle tyre either.
